FDA Approves New Biologic Treatment for Wet Age-Related Macular Degeneration

Published: Fri Jun 30 00:00:00 +0000 2006

The Food and Drug Administration (FDA) today approved Lucentis (ranibizumab injection) for the treatment of patients with neovascular (wet) age-related macular degeneration (AMD). Lucentis is the first treatment which, when dosed monthly, can maintain the vision of more than 90 percent of patients with this type of AMD. Lucentis is a new molecular entity (NME), meaning it contains an active substance that has never before been approved for marketing in any form in the United States. Lucentis will be the first FDA--approved product to provide prescription information in the new format for prescription drug package inserts, to provide professionals and consumers clear and concise prescription information.

"This approval is of great importance for the 155,000 Americans who are diagnosed each year with AMD, a common cause of severe and irreversible vision loss in older adults," said Dr. Andrew von Eschenbach, Acting Commissioner of Food and Drugs. "At a time when our elderly population is rapidly increasing, this product preserves quality of life for those affected by this disease, helping them to regain the ability to participate in everyday activities such as reading and driving."

AMD, a retinal disease causing severe and irreversible vision loss, is a major cause of blindness in individuals older than 55 years. Untreated, the majority of eyes affected with wet AMD may become functionally impaired. Wet AMD, which accounts for 10 percent of all AMD, is responsible for 80 percent of the associated vision loss.

The vision loss in wet AMD is caused by the growth of abnormal leaky blood vessels that eventually damage the area of the eye responsible for central vision. Lucentis is designed to block new blood vessel growth and leakiness, which ultimately lead to disease progression and such vision loss.

Lucentis, a biologic product, administered by injection into the eye, was shown to be safe and clinically effective in three multicenter, randomized studies of patients representative of the population usually affected with AMD. In clinical trials, nearly 95 percent of the participants who received a monthly injection maintained their vision at 12 months compared to approximately 60 percent of patients who received the control treatment.

Approximately one-third of patients in these trials had improved vision at 12 months. In a single study carried out for 24 months, these findings have been maintained with continued monthly dosing. The most commonly reported adverse events included conjunctival hemorrhage, eye pain, floaters, increased eye pressure and inflammation of the eye.  Serious adverse events were rare and often related to the injection procedure including endophthalmitis (severe inflammation of the interior of the eye), intraocular inflammation, retinal detachment, retinal tear, increased eye pressure and traumatic cataract.

Lucentis is manufactured by Genentech, Inc. in South San Francisco, California.

During the first 6 months, I experienced night sweats, water retention, general malaise. From months 6-9, I gained 24 lbs. I am a vegetarian who works out 4-5 days a week. I finally read a blog where women were complaining of the same issues. My GYN told me that it was just my body adjusting. Had it removed in July. Lost 11 lbs the first week and then stalled. Had some blood work done. It revealed that my Thyroid function was practically non-existent. I am now on Thyroid medication and I have finally lost the last of the 24 lbs. No more Mirena and said "buh bye" to my GYN.

Abilify caused DYSTONIA, chills, cold sweats, sweating, and anxiety. I've obviously stopped taking this poison (in MY case) but today is my first day stopping. Hopefully my symptoms will clear up in a few days. I've only been taking the medicine for right at 11 days. I've just returned from the ER where they have said there's really nothing they can do and I have to "wait it out." Nice.

I had my Mirena removed on Thursday, September 2nd 2010. Today is day 3 without that monster. I have yet to start bleeding. My swelling in my stomach has done down considerably. I can hardly believe how much flatter it is. UNREAL! OH and my libido is super duper high!!! Holy hanna! I feel like a teenager again. I am still a bit dizzy and emotional...but I am certain this is from my hormones slowly going back to normal. I KNOW taking the Mirena out was the best decision for my body, mind and spirit. I will keep everyone posted in the days to come. So far I am THRILLED with just the way my body looks and feels. Carol

My neurologist prescribed Topiramate for facial pain. I took four pills-1/day at bedtime starting 7/16/10. I went off the pill 7/20/10 because I was not having a good reaction to it. My headache started Sunday evening 7/25/10. My distance vision was blurred, and my near vision perfect. This was totally opposite of what my vision should be. I had a terrible headache and took 3 aspirin and went to bed. I woke up Monday, 7/26/10 7:00am and I could not see. I had a terrible headache. I went to emergency room, had a catscan, bloodwork and was told to see my opthamologist immediately. I went to the doctor who, thank goodness, was only 1/2 mile away. She saw me immediately and said she had never seen this before-my ocular pressure was so high it had distorted the shape and caused the vision changes.. She left the office and came back with eyedrops to drop the pressure. She told me I would have been blind had I not arrived at the office when I did. We did 3 different drops and pills for 4 days. I saw the doctor every morning and slept for 4 days and could not drive. My vision in left eye is still not sharp. My night vision was razor sharp-now it is blurred. I am still taking a drop for the swelling in the back of my left eye. It took 2 weeks before I could read for more than 15 minutes. I still(9/5/10) have pressure in back of the left eye and my left eye vision is still not back to normal. My readings are all fine, but the pressure is still felt. Anita Rosner
