I've been searching for answers on LONG TERM side effects of Lupron hoping that it could give me answers as to what's going on in my body now!
23 yrs ago my fertility dr. gave me Lupron to inject MYSELF EVERY DAY (experimental back then). I had SEVERE endo since the age of 16 and was fighting that as well as trying to get pregnant thru invetro. I was 26. At 29 I had an emergency hysto. Now at 48plus I have chronic tendonitis where the tendons will shred off of the joints. I have been diagnosed with C spine and T spine disease, osteoarthritis and a mystery that I'm going to have to go to the Mayo to figure out (hopefully). I have tumors that are growing in my lower back...growing around nerves, tendons and bone, They aren't sure why they are there, why they are growing, or even what they are. Surgery removed 2 of them...but pathology shows foreign matter other than tissue/bone. All they DO know is that it is not cancer. One year ago I started losing weight (25 pds) for no reason. I'm now holding my own at 112 and hoping to figure all of this out. I can't help but wonder if Lupron or any other experimental drugs I was on has something to do with this. Is there anyone out there that used this drug so long ago??

Researchers have been doing studies for many years regarding trying to determine the role of genetic factors in patients response to Singulair (Montelukast).

This study from Spain identified the following gene variations hypothesize to be related to leukotriene pathway response. Sixty one patients with asthma were studied. Three gene types were identified:

type 1. Thirty-two patients (52.5%) were homozygous for the five repeats allele;
type 2. 17 (27.9%) were heterozygous (4/5 repeats)
type 3. 12 (19.7%) were homozygous for 4/4 repeats.

The study showed that montelukast was effective for types 1 and 2 but not effective for type 3. Type 3 represented approximately 20% of the group study.

"After the montelukast treatment decrease number of asthma exacerbations, improvement of FEV(1) and decreased use of beta(2) agonists was observed in patients with 5/5 or 4/5 repeats. Conversely, the patients with 4/4 repeats genotype did not modify these data after treatment."

So it seems logical that if it can be identified that montelukast is not effective for certain gene type variations, then montelukast could cause adverse side effects in certain gene type variations.

It is interesting that 20% of this group does not respond positively to montelukast. That is the exact same number that even Merck says gets a headache from montelukast. Headache is the highest incidence of adverse side effects that has been reported. That comparison, however, is just a coincidence because it has not been studied and proven. Maybe.

Where are the studies that pertain to gene type variations and adverse side effects? You would think that somebody could do them.

BACKGROUND: It was hypothesized that asthmatic patients with mutant alleles in the leukotriene pathway should not respond to leukotriene receptor antagonists and the concept of a tailored treatment is increasingly supported. METHODS: Sixty-one patients (mean age 24.9 years, range 14-52) with moderate persistent asthma were clinical and immunological assess prior and after a 6-month treatment with montelukast. Tandem repeat polymorphisms were genotyped in the promoter (-147 to -176) of 5-lipoxygenase gene (ALOX5). RESULTS: Thirty-two patients (52.5%) were homozygous for the five repeats allele; 17 (27.9%) were heterozygous (4/5 repeats) and 12 (19.7%) were homozygous for 4/4 repeats. After the montelukast treatment decrease number of asthma exacerbations, improvement of FEV(1) and decreased use of beta(2) agonists was observed in patients with 5/5 or 4/5 repeats. Conversely, the patients with 4/4 repeats genotype did not modify these data after treatment. CONCLUSIONS: It was confirmed that ALOX5 promoter polymorphisms have a clear influence in montelukast response in atopic moderate persistent asthma patients. The genetic study could identify those patients most likely to respond to montelukast.

extremely depressed after a week of taking levoxyl. Also face is swollen, eye swollen and red and face and eyes burn. These were the same symptoms after taking synthroid after a week. What's next? I am close to being suicidal after having survived lung cancer, surgery, chemo and radiation. To get another primary cancer and have things slide downhill after such a remarkable recovery, I can do nothing but cry after making the decision to have a cancerous thyroid removed that could have served me for years and years since it was such a slow growing cancer. I should have followed my instincts and just left it alone. Now all I want to do is lie on the couch and cry.

I've been on Synthroid for 8 months after gaining 10 pounds in two months. The symptoms started to appear when my Mom was very ill. Doctor said it was caused by stress and started me on 25 mcg of Synthroid. My reading at that time was 5.56. I had blood work done every 4 weeks,and the numbers are going down (1.8 last reading), but so is the quantity of my hair, which I see all over my white tile. My weight hasn't budged even though I am dieting. My doctor upped me to 100 mcgs. My blood work is not good - low white count, low lymphocytes, borderline anemic. Mom died, and I didn't get any better. I feel sick all the time and it's difficult for my husband to understand how bad I feel. I've tried to get my doctor to prescrive Armour, but he doesn't even return my phone calls. When I see him in the office, he keeps telling me it takes time. I hate living this way and want to stop the pills completely.
