We have all been saying that our issues regard not being informed about all of the possible side effects. And, we know that Singulair works well for some people. Nobody wants to take a good drug away from those for which it probably performs miracles. People who have toxic side effects have a right to know up front. My observations about montelukast's chemical structure are either general or not quite 100% correct or could be quite vague - so forgive me. I do not claim to be good at organic chemistry. But from doing a little work, I have come up with some observations. 1. It would seem to me that montelukast might work quite well for people who have developed mold category related asthma. I observed that chloroquinolin, a component of montelukast, is a good fungicide effective against Aspergillus, Alternaria, Cladosporium, Penicillium and Candida. Dust mites can only digest if helped by aspergillus so they go into the mold category. Molds produce millions of spores so anyone who lives in contact with mold would be chronically sick from their presence. Then people get hypersensitized to that. I am probably wrong but I could imagine that montelukast is: 1) a ligand that binds to an empty cysLT1 receptor for a period of time 2) 7-chloroquinolin-2-yl which either acts intact or breaks down into a quinoline fungicide so that it kills the chronic mold/fungus infection and 3) a sulphur/methyl anti-inflammatory component that tells the t-cells that they are not needed so they will die. Wow, that would be great for mold asthma if it was completely non-toxic. It would be also great under controlled circumstances for many people who are mold-miserable. If I am wrong, I better go out into my garage and start inventing such a drug. This is my visualization to try to explain the side effects of neurotoxicity. So adverse reactions could be to the quinoline component as an allergic reaction or dose related so that it just built up to a toxic level over time. There are many signs that t-cell populations are significantly reduced by montelukast. The fact that the Italians can do it in the test tube might be that it's a chemical component of montelukast designed to cause the t-cells to die. Montelukast is a large molecule so Artie says it cannot penetrate the blood brain barrier. That would be an argument if nobody was complaining about neuro-psychiatric side effects. The neuro-psychiatric side effects are identical to quinoline and quinolones. When I read about Lariam, it just sounds like a more extreme version of Singulair side effects. Chloroquinolins were used before they invented Lariam, which is stronger. The malaria Plasmodiums became immune. Hallucinations, anxiety, depression, suicidal thoughts are completely consistent in all of the quinoline/quinolones. If montelukast breaks into sub-molecules then quinolines easily penetrate the blood brain barrier. I find clinical evidence that montelukast may act as more than more molecule. And, that there is a rational for the existence of the chloroquinolin and evidence that it may be the source of toxicity. I am glad to risk being called crazy. That is what the internet is for. We can present our ideas and discuss. So, just take this with a grain of salt. If I am close to the truth, this post will find it's proper home.

Actually, I am of an age when many of my friends are getting cancer. Cancer is a baffling issue. Leukotrienes are being studied as an anti-cancer category because some cancer tumors express leukotriene receptors. One of my friends has prostate cancer that expresses the cysLT1 receptor. What the heck is that? Is it a mast cell that went bad?

That is why I say that leukotrienes are the twilight zone of the immune system. What is the cause and what is the effect? Why does it go in a circle?

I think that it all means that balance in the immune system is very delicate. The entire system has to work together. That is how I ended up here and interested in Singulair. I was really researching leukotrienes and cancer.

Something bothers you about my interest in Singulair enough to ask. Take what I say, if you care to read it, and analyze it for yourself. If it encourages you to research for yourself, that would be good. Don't take my opinion as anything more than my ideas. I don't have answers.

I'll leave the research to the experts - not that I'll give them my trust. I ask a lot of questions too. I just don't want people w/o a medical/research background to get confused about fact vs. speculation.

I also read your latest post on Yahoo implying a cancer risk in children. That seems likely to scare people w/o much evidence. We have enough to deal with in the shorter term w/o adding that. I'm not saying what you're doing is intentional; it just seems that you've gone way beyond the available "facts".

What do you want of people who know that you can only push nature so far? Facts come after the fact. Who is going to warn? Nobody. It is a long term process that the immune system is attacked over and over to cause cancer. Some times we cannot prevent it. If you think that I have some kind of motive to warn, you are mistaken.

By the way, who are experts? If the leading expert spoke anonymously, would we listen? Do we listen to the words or the authority? You decide. I made my decision long ago.

Things change in science. Ideas always change. Some people think that dopamine is not infuenced by quinolines. Well, wrong to that also. I am not coming back.

Why are you being so sensitive? I'm not insulting you or your motives. I just wanted you to consider how people will be affected by your exploration. I am a skeptic, but when people participate in these sites, they are often feeling desperate and vulnerable.

Absolutely, there is NO freakin' way that people are experiencing these neurological side effects without a very, very good reason. Maybe that reason was built into the original research which had a different purpose than some pill that was supposed to be for everybody with a cough, swollen sinus, or a sneeze. How does the original plan to identifiy what causes asthma and design a medication that solves an awful problem go to a medication that gets prescribed for seasonal allergies - pollen? Not even, the same category.

I give up. Chloroquinolins?? Maybe somebody will explain without sweeping this under the rug.

Yes some of the info you have posted can be scary when thought about. But I for one want to know about it. There may be long lasting effects we don't know about. What you have provided was documented studies & information (including links). Yes, you would give us your opinion and you always clearly stated it was your interpertation that that we should speak to our Dr's. Yes, this is a world that will have many opinions and they all can be heard. Some people may not want to hear everything. It is their decision whether to read your posts or not. I appreciate your posts both here and on Yahoo and I am thankful for the time and effort you have put in to making us aware of the info that you have found out. I hope you reconsider your Yahoo membership as you will be missed.
